In the yeast genome the gene for the human androgen receptor (hAR) and a reporter gene cassette have been integrated.
Androgens and androgenically active substances bind to the receptors and the resulting complexes trigger the transcription of the reporter gene.
The reference standard for calibration and quantification is 5α-Dihydrotestosterone (DHT).The dose-response curve corresponds to a non-linear model.
The detection is performed photometrical after reaction of the reporter enzyme with the chromogenic substrate p-nitrophenyl phosphate at 405 nm.
