Product Description
The S-YTSβ is an effect-based 96well assay for the the determination of thyroid receptor β mediated effects expressed as T3EQs (triiodothyronine equivalent concentration) in aqueous samples by means of the yeast Saccharomyces cerevisiae*. No sterile conditions are required for the whole test procedure.

Test Principle

Genetic Modifications

This yeast strain contains the gene for the human thyroid receptor β, the gene for the steroid receptor coactivator 1e (src1e), specific gene regulation sequences and the reporter gene lacZ.

Transactivation

Thyroid active substances bind to the thyroid receptors and the resulting homodimers trigger the transcription of the reporter gene.

Calibration

The reference standard for calibration and quantification is triiodothyronine (T3). The dose-response curve corresponds to a non-linear model.

Detection

The detection is performed after reaction of the reporter enzyme β-galactosidase with the chromogenic substrate CPRG at 580 nm.
